Svar Life Science: Complement System Solutions
The complement system is an integral part of the innate immune response. Given its important role in many human disorders, complement activation studies are important in a wide range of applications – from drug development and medical device safety to clinical diagnostics and basic research. There are three pathways of complement activation: the classical pathway, which is triggered directly by the pathogen or indirectly by antibody binding to the pathogen surface; the MB-lectin pathway; and the alternative pathway, which also provides an amplification loop for the other two pathways.
At the basic level, the broad functions of the complement system can be split into three areas: (1) the activation of inflammation; (2) the opsonization (labeling) of pathogens and cells for clearance/ destruction; and (3) the direct killing of target cells/microbes by lysis. C3 and C4 are the most commonly measured complement components. A complement test may be used to monitor people with an autoimmune disorder. It is done to see if treatment for their condition is working. When the complement system is turned on during inflammation, levels of complement proteins may go down.

Based on the well-established and robust ELISA technology, our products can be used in standard setups as well as automated setups with ELISA robots. Functional Assessment
The Svar functional complement assays under the Wieslab® brand were developed together with a network of global key opinion leaders (KOLs) in 2005 and have since been used to answer the need for an easy-to-use method of detecting complement deficiencies and to explore the effect of complement targeting drugs. The Alternative Pathway amplification loop is central to the essential amplification of the complement response. The novel, Svar Factor P functional assay has been developed to enable analysis of the complement up-regulator, Properdin, and its associated C3-convertase function.
The Svar complement functional assay solution is an ELISA-based platform that enables the determination of specific activity of all three pathways separately without interference from the others. The well-established ELISA technology means a robust and standardised assay platform, delivering fast, reliable, and objective results that are easy to interpret within as little as 3 hours.

cell-based complement assays offer a new way of studying the complement system. Based on iLite® technology, these assays use a reporter gene system to detect the presence of the complement component for which they were designed. iLite assays offer excellent sensitivity and specificity which ensures that you get accurate results. As an added convenience, all iLite cell lines come assay-ready – no cell culturing is required. This results in a faster workflow and dramatically improved reproducibility and overall performance.
iLite cells express receptors on their cell surfaces that are specific for the complement component for which they were designed. When a complement component binds to its receptor, an intracellular signaling cascade is initiated which will result in the expression of luciferase. The amount of luciferase generated can be used to calculate the amount of the complement component in the sample. Many iLite assays also include a second constitutively expressed luciferase gene which can be used for normalization.
The purpose of the Complement TCC Flexi reagents is to specifically detect the soluble terminal complement complex (TCC, also known as sTCC or sC5b-9) which is an end product of complement activation of all three complement pathways. The Complement TCC Flexi application note and reagents enables users to independently set up, customize and optimize a Complement TCC Flexi assay based on Svar Life Science’s Complement TCC assay on Gyrolab® (Gyros Protein Technologies) automated platforms or other platforms of their choice.

Complement Flexi- for Automated Platforms
The Svar Life Science Complement Flexi Solutions allow users to independently set up, customize and optimize assays based on existing Svar Life Science assays. In collaboration with Gyros Protein Technologies, Svar has developed application notes for use of the Complement Flexi Solutions on the automated Gyrolab® platforms. The Flexi Solutions can of course also be used in manual assays as well as for other laboratory techniques, such as immunoblotting and immunohistochemistry, where your imagination is the only limit. Complement Activity Biomarkers
The Svar Complement activity biomarkers give valuable intelligence in several situations where one might suspect that complement activation plays a role in the disorder. Complement TCC
The Terminal Complement Complex, or TCC, is a product of the terminal pathway and thus a result of activation of any of the three complement activation pathways. As a reflection of total in vivo complement activity, increased levels of TCC can be detected in several acute, flaring, and chronic diseases.
Complement C4d
C4d is a marker for complement activity originating from the classical or lectin pathways. It is an established histological biomarker with a strong association to antibody mediated rejection of grafts. Moreover, C4d has increasingly been used as a biomarker for diagnostic and prognostic purposes, in conditions ranging antibody-mediated disease to cancers.
Complement Factor P
Factor P, also known as Properdin, is the only known positive regulator of alternative pathways of complement response and the key to ensuring sufficient complement activity. Properdin imbalance or dysfunction has been linked to several health disorders, and assessing its prevalence is a useful tool for a better understanding of complement regulation. In addition to the quantitative Factor P assay linked below, we also offer a functional Factor P assay.
Complement Testing Application:
