Proteolysis targeting chimeras (PROTACs) are heterobifunctional small molecules with three chemical elements: a ligand binding to a target protein, a ligand binding to E3 ubiquitin ligase, and a linker for conjugating these two ligands. It is a chemical knockdown strategy that degrades the target protein via ubiquitin-proteasome system. PROTAC's have been found to be important in the following areas: Overcoming drug resistance of cancer, Eliminating of both enzymatic and non enzymatic functions of the kinases, degrade the "undruggable" protein target, fast and reversible chemical knockdown strategy in vivo.
