| Scientific Background | The inhibition of kallikrein 5 (KLK5) has been identified as a potential strategy for treatment of the genetic skin disorder Netherton syndrome, in which loss-of-function mutations in the SPINK5 gene lead to down-regulation of the endogenous inhibitor LEKTI-1 and profound skin-barrier defects with severe allergic manifestations. To aid in the development of a medicine for this target, an X-ray crystallographic system was developed to facilitate fragment-guided chemistry and knowledge-based drug-discovery approaches. |
| Molecular Mass | The protein has a predicted MW of 32.51 kDa. Due to glycosylation, the protein migrates to 40-50 kDa based on Bis-Tris PAGE result. |
| Sl Sequence | Ile67-Ser293 |
| Solubility | Dissolve the lyophilized protein in 50mM MES, 150mM NaCl (pH 5.5). It is recommended to resuspend at 0.5 mg/mL if the lyophilized powder is 100 μg or less, at 1mg/ml for 500ug or 1mg lyophilized powder. Do not mix by vortex or vigorous shaking.
