Primary Antibody
A primary antibody plays an important role in the detection of biomarkers for diseases such as cancer, diabetes, Parkinson's, and Alzheimer’s disease, as well as in the study of absorption, distribution, metabolism, and excretion (ADME) and multi-drug resistance (MDR) of therapeutic agents.
Secondary Antibody
Secondary antibodies provide signal detection and amplification, along with extending the utility of an antibody through conjugation to proteins. Secondary antibodies are especially efficient in immunolabeling. Primary antibodies are linked to the target antigen directly by secondary antibodies (s). In immunolabeling, the Fab domain of the primary antibody attaches to an antigen and releases the secondary antibody's Fc domain.
Secondary antibodies can be conjugated to a variety of molecules, including fluorescent dyes like fluorescein isothiocyanate (FITC), rhodamine derivatives, and Alexa Fluor dyes, as well as enzymes like horseradish peroxidase (HRP) or alkaline phosphatase, in order to be used in a variety of applications (AP). Secondary antibodies are used in many biochemical assays, including:
- ELISA, including many HIV tests
- Western blot
- Immunostaining
- Immunohistochemistry
- Immunocytochemistry
Isotype Control
Isotype controls are primary antibodies that are similar to the class and type of the primary antibody employed in the application but lack specificity for the target. To assist in separating specific antibody signals from non-specific background signals, isotype controls are used as negative controls.
Proteins and Peptides
Proteins and peptides are fundamental components of cells that carry out important biological functions. Proteins give cells their shape, for example, and they respond to signals transmitted from the extracellular environment. The regulation of other biomolecules is significantly controlled by specific types of peptides. Proteins and peptides share a similar structural makeup in that they are composed of chains of amino acids that are joined by peptide bonds (also called amide bonds).
ELISA Kits
ELISA, also known as enzyme-linked immunosorbent assay, is the basic technique that is carried out to detect and quantify antibodies, proteins, peptides, and hormones in the sample, like blood, serum, plasma, and tissue. Antibodies are the proteins produced in blood in response to the immunogenicity illicit by the specific antigens.
It helps in the examination of the presence of antibodies in the body in response to certain infectious diseases. ELISA is a unique analysis in comparison to other antibody-assays as it gives qualitative and quantitative results together with the separation of specific and non-specific interactions taking place via serial binding to solid surfaces, which is typically a polystyrene microtiter plate.
Assay Kits
Bioassays are ideal for determining drug potency and stability using a simple, mix-&-read protocol that reflects the drug’s clinical mechanism of action. These rapid cell-based assays bypass the need for expensive and time-consuming cell culture, reducing lab work errors. Additionally, bioassays decrease assay development and validation times, provide accurate and precise data, reduce costs, facilitate a simplified global method transfer, and ultimately accelerate potency testing and QC lot release.
RNA Interference
RNA interference (RNAi) is a genetic regulatory system that functions to silence the activity of specific genes. RNAi occurs naturally, through the production of nuclear-encoded pre-microRNA (pre-miRNA), and can be induced experimentally, using short segments of synthetic double-stranded RNA (dsRNA).
