CAR- T Cell therapy
Chimeric Antigen Receptor (CARs) are the natural receptors that provide unique properties to T cells for attacking specific targeting proteins/antigens or cancerous cells. CAR T cell therapy has emerged as a novel therapeutic technique that now appears as a revolutionary therapeutic approach in cancer treatment. The significant benefit of this therapy is that tumour antigen is directly identified without the need for a major histocompatibility complex for antigen expression. Engineering T cells to boost the immune system has potential benefits beyond cancer treatment, including treatments for HIV/AIDS, chronic inflammatory disease, arthritis, and lupus. BPS has developed stable, recombinant cell lines expressing cancer antigens at different expression levels--high, medium, or low expression. These cell lines are ideal to monitor the effectiveness of different CAR-T at detecting the target antigen expressed at different levels, which may relate to the time dependence of antigen expression. BPS also offers biotin-labeled versions of many of the antigens recognized by CAR-T cells, such as BCMA, CD19, CD22, CD123, CD38, and ROR1.
CRISPR-Cas9 is a tool used for genome editing that has been adapted from the bacteria’s innate immune response. Bacteria capture and store DNA fragments from invading viruses or DNA within a region of their genome termed the CRISPR (Clustered Regularly Interspaced Short Palindromic Repeats) array. This region is continually transcribed, producing short CRISPR RNA (crRNA) guide sequences that, when they detect an invading virus or DNA whose sequence is complementary to it, bind and recruit the Cas9 (CRISPR-associated protein 9) nuclease, which then specifically cleaves the invading DNA. This results in degradation of the invading DNA, thereby protecting the cell. There are seven known human coronaviruses, including the novel SARS-CoV-2. Four of these strains cause only mild respiratory symptoms. The SARS-CoV-2 virus consists of four different proteins and a strand of RNA. The most prominent feature, a trimer formed by the “Spike” protein, sticks out from the membrane and gives the virus its distinctive “corona” structure. Two other proteins, envelope protein and membrane protein, reside in the membrane between these spikes, providing structural integrity.
