BirA Biotinylation Kit
BirA, also known as biotin ligase, biotin operon repressor protein, biotin holoenzyme synthetase, and biotin-[acetyl-CoA carboxylase] synthetase, has a molecular weight of approximately 35 kDa. Biotin-labeling is commonly used for non-radioactive labelling and purification of proteins and other target molecules. Biotin labelling takes advantage of the exceptionally strong interaction between biotin (vitamin H) and either avidin or streptavidin. The affinity of biotin for avidin or streptavidin is one of the strongest known non-covalent interactions of a protein and ligand, exhibiting a dissociation constant (Kd) of around 15–20. BirA protein ligase covalently adds biotin to biotin-acceptor peptides containing an avidin sequence in a highly efficient and specific manner in a reaction that requires ATP.
