OC2-K3-E10 LNPs delivered Firefly luciferase mRNA and resulted in comparable levels of luminescence as SM-102 LNPs. These LNPs also successfully delivered Cas9 mRNA and sgRNA. LNPs containing OC2-K3-E10 were able to encapsulate four-fold more mRNA copies per particle compared to SM-102.
ZA3-Ep10 is a zwitterionic amino lipid (ZAL) that is uniquely able to deliver long RNAs including Cas mRNA and sgRNA. ZAL nanoparticles' delivery of sgRNA enables permanent DNA editing. ZNP delivery of mRNA represents a promising step towards improving the safety and utility of gene editing.

LP-01 is an ionizable cationic lipid with a similar structure as 11-A-M (Echelon Cat#: N-1111). LP-01 can form lipid nanoparticles (LNPs) with cholesterol, DSPC, and PEG2k-DMG. The LNP nucleic acid assemblies can deliver single-guide RNA (sgRNA) targeting MHC class II trans-activator (CIITA) in T cells.
PPZ-A10 is an ionizable lipid used for formulating lipid nanoparticles to deliver RNA in vitro and in vivo. PPZ-A10 LNPs preferentially delivered Cre mRNA to liver and spleen immune cells at concentrations as low as 0.3 mg/kg. Decreased tumor burden and increased survival was observed in hepatocellular carcinoma mouse model treated with PPZ A10 LNPs.
